Over the past year I have tried various inexpensive telomerase activators on my scalp such as fenugreek extract, black cohosh extract, and colostrum solution. However, none of these seemed to work as well as more expensive astragalus extract previously tested to restore hair color, although astragalus extract takes about a year to do the job. So for these recent pictures I dyed my hair prior to returning to astragalus extract treatment of my scalp. The earlier before and after pictures of my hair using astralus extract did not include any hair dye.

Inhibiting Cellular Senescence with Telomerase Activators
During February and March 2013 I threw in more Milk Thistle Extract (10 grams/day), and added Hawthorn (5 grams/day), both of them telomerase activators suggested by Product B. I dropped the HGH-elevating alpha GPC to finance that, however. I got sugarless creatine monohydrate this month for more ATP to supply to adenylate cyclase from exercise to create cyclic AMP for inhibiting caveolin-1 to restore senescent cells.

At this time my approach here is to use a number of telomerase activators [List 153] in parallel as I attempt to meet the original goals using cyclic telomerase activation, two weeks of telomerase activators followed by two weeks of telomerase inhibitors with anticancer neutrceuticals. Fourteen endogenous telomerase activators from exercise may be amplified with specific secretagogue supplements, and I am still using astragalus extracts and astragalosides, in addition to Fenugreek extract, ginkgo biloba ginkgolides, and nitric oxide boosters. The mainstay of the 2-week telomerase activation phase in March 2013 was astragalus root at 33 grams/day, astragalus extract for the scalp (5 droppers/day of 30 drops/milligram of astragalosides), with 4 x (175 mg/cap) capsules/day of black cohosh extract taken orally, 3.25 grams/day of Fenugreek extract (5 x 620 mg taken orally before bedtime) for diosgenin and HIF-1 transcriptional activation of hTERT, colostrum taken orally (4 x 500 mg/day) and as a skin cream (6 x 500 mg/day in 1/6 cup water), arginine (8 to 10 grams/day) for nitric oxide telomerase activation in endothelial progenitor cells, DHEA (4 x 25 mg/day) for testosterone [List], Alpha GPC (1-2 grams/day) for supplemental HGH activation of hTERT, and ginkgo biloba (4 grams/day) for HIF-1 hTERT transcriptional activation. In addition, I used moderate exercise and associated supplements including arginine and alpha-GPC to boost the 14 exercise-induced telomerase activators. Also note that antioxidants such as vitamin C inhibit telomere shortening leading to premature stress-induced senescence, as do homocysteine blockers.

Recovering from Cellular Senescence
Exercise also boosts cyclic AMP, which can cause a senescent cell to recover its youthful phenotype by inhibiting caveolin-1, restart its cell cycle, and lengthen its telomeres in response to further stimulation from EGF or PDGF, from exercise, colostrum, or other supplementation. The generation of cyclic AMP works best when ATP is abundant from creatine monohydrate. Stimulating the PI3K/AKT pathway with folic acid (vitamin B9), exercise and supplements also sequesters FOXO factors away from the cellular nucleus, reducing the expression of caveolin-1 (gene CAV1) associated with the transition to the senescent state of the cell and the initiation of senescent growth arrest when this internalizes cell membrane caveolae, blocking communications via growth factors such as EGF and PDGF. In addition, exercise reduces P16INK4A levels, which can also be lowered with carrots and retinol. P16INK4A makes recovery from cellular senescence difficult, but can be dramatically reduced, as it is a ubiquitinated protein that can also be downregulated.

Exercise can be continued if high levels of vitamin K2 (from Swiss cheese or MK-7 supplements) and vitamin D are used to stop calcification of the arterial system associated with aortic stenosis. To continue exercise it is also desirable to take B-vitamin homocysteine blockers to stop the development of atherosclerotic plaque.

2013 Life Extension Program

I used astragalus extract on my scalp again, but added tribulus, which promotes the expression of melanocyte-stimulating hormone, and took ashwagandha, which elevates endogenous antioxidants including catalase. (In July 2012 I substituted bacopa for ashwagandha, which also boosts the endogenous antioxidants. However, bacopa elevates thyroid hormone T4, and I took it 1-2 grams with meals for a while to improve my leanness. Furthermore, some vendors now claim that bacopa is a telomerase activator, although this is not now well documented in publicly available scientific studies.) Catalase is believed to inhibit the bleaching of hair from the inside out with endogenous hydrogen peroxide. Melanocyte-stimulating hormone from tribulus is believed to darken hair. Instead of applying Sequel Rejuvenating Colostrum Skin Cream, I mixed up a slurry of bovine colostrum in water and rubbed it in like a skin cream. Colostrum contains TGF-beta, a telomerase inhibitor that reconstructs the extracellular matrix including both collagen and elastin. It also contains many other telomerase-activating growth factors found in growth factor skin creams including TGF-alpha, EGF, IGF-1, IGF-2, VEGF, FGF, and PDGF. In addition to mixing NOW Colostrum (powdered) with water to apply it as a skin cream, I have also tried mixing it in olive oil, in which colostrum is not soluble, however. Both formulae may be useful, but the solubility of colostrum in water makes the aqueous formulation most attractive. Perhaps a paste of powdered colostrum and water would be effective applied as a mask. Colostrum water may be helpful applied to the scalp.

2013 Recommended Life Extension Program

I was formerly sometimes using bacopa to upregulate endogenous antioxidants including SOD, catalase, and glutathione peroxidase, before reading that some vendors claim bacopa as a telomerase activator. In June 2011 I used ashwagandha [withania somnifera] to upregulate SOD, catalase, and glutathione, relying on melatonin to upregulate glutathione peroxidase during the last half of the month. Catalase upregulation should be useful to oppose hair greying.

For telomerase upregulation I took 33 g of astragalus root/day in March 2013 with astragalus extract including 10 mg of astragalosides. I upregulated nerve growth factor with acetyl L-carnitine (4 x 400 mg) and alpha lipoic acid (4 x 200 mg), which promotes Id-1 helix-loop-helix transcription factor, which upregulates the production of hTERT mRNA. In addition, I took rosemary tea, which also upregulates nerve growth factor and Id-1. To activate telomerase in endothelial progenitor cells I took Ginkgo Biloba (1.5 grams/dose x 4 doses), which upregulates HIF-1 transcription factor, and L-arginine (10 grams), which upregulates Nitric Oxide. HGH was upregulated with whey protein and 1.5 grams of arginine plus 1.5 grams of leucine. In July 2012 I used alpha-GPC as an HGH secretagogue for boosting hTERT mRNA levels with HGH. After evening exercise, I took 620 mg x 5 = 3.25 grams of Fenugreek extract to upregulate insulin and phosphorylate tankyrase 1 to remove the TRF1 loop closure protein to open telomere t-loops for access by the telomerase holoenzyme. Fenugreek also upregulates testosterone, a telomerase activator in for cardiac cells, muscles and melanocytes, and provides diogenin, another HIF-1 transcription factor upregulator. At this low dose, no undesired transformations are expected from Fenugreek phytoestrogens when the application period is just two weeks.

Upgrades for 2013

Cycloastragenol and Astragalus Extract for Primary Telomerase Activation
In April 2011 I used experimenter's cycloastragenol (10 mg/day) from a chemical supply company for the rejuvenation application for the first 15 days of the month, plus a couple of bottles of GAIA Herbs astragalus root extract in glycerin (green label, 1 mg astragalosides per 30 drops), which I applied to my scalp. I tried in 2011 to put the cycloastragenol in via a transdermal route of application scheme. By July 2012 I concluded from studies done by the VIDA Institute, Terraternal, and Calvin B. Harley, et al., that astragalus root at 25-33 grams/day would be more effective than cycloastragenol (25 mg/day), TA-65 (25 mg/day), or astragaloside IV (100 mg/day) for lengthening telomeres to obtain rejuvenation effects. I tried shelling astragalus root powder out of gelatin capsules, mixing it as a suspension in water, and heating it up to be served as a tea with sweetener. Although you have to stir the mixture with a spoon because it somewhat settles out of solution, this may be easier than swallowing 66 x 500 mg capsules throughout the day. I finally settled in March 2013 for 4 doses of astragalus root pills during the day, 9 grams + 8 grams + 8 grams + 8 grams = 33 grams = 18 caps + 16 caps + 16 caps + 16 caps.

Forskolin & Tribulus for Hair Darkening Improvements
I started taking Forskolin to darken my hair toward the end of the month in 2011, after I read that Forskolin darkens hair, and upgraded my Hair Greying index entry on the topic. Forskolin also elevates the expression of testosterone, which promotes reverse cholesterol transport and activates telomerase in muscle tissues and hair follicles via the androgen receptor. Tribulus, an aphrodisiac, also darkens hair by increasing the expression of melanocyte-stimulating hormone.

Thymic Restoration with Telomerase Activation from Nitric Oxide
In April 2011 and July 2012 I used 10 grams of arginine per day during the first 15 days of the month in order to stimulate the thymus gland, restore youthful levels of thymic hormones, and rejuvenate endothelial progenitor cells.

Confining High Vitamin D to Telomerase Inhibition Periods
Otherwise, I followed the routine from the prior month, except that I used high vitamin D levels only during the final 15-day telomerase inhibition periods, as I confirmed in February that Vitamin D3 is a telomerase inhibitor.

Nicotinamide to Improve the Hayflick Limit
I tried again to follow Kang's advice on nicotinamide [Links], which has extended life spans of cells in the lab by a factor of 1.6. (See Kang HT, Lee HI, Hwang ES (2006), Nicotinamide extends replicative lifespan of Human Cells, Aging Cell, 5(5): 423-36.) This is due to fewer telomeric repeats lost per cell division, so that cells can divide more. In April 2011 I could obtain 500 mg capsules of Now Foods nicotinamide (niacinamide, a form of niacin, also a form of Vitamin B3), which I added to my usual 4 daily Super B-Complex pills including 100 mg of niacinamide [Links, also called nicotinamide], in addition to other B-vitamins and material functioning like a homocysteine shield against atherosclerotic plaque. I took the nicotinamide recommended by Kang, 500 mg in the morning and 500 mg before bedtime.

Nicotinamide to Increase Tankyrase Expression and NAD+ levels for opening T-Loops
I note that my recent research shows that niacinamide can be taken < 3 grams/day (while taking a telomerase activator) to provide the NAD+ substrate required by tankyrase 1 to strip TRF1 telomere binding protein with poly(ADP-ribosylation), opening telomere loops for telomerase access and subsequent extension of the telomere. Alternatively, NAD [Index] can be taken directly as a supplement to enhance TRF1 t-loop closure protein stripping. Furthermore, insulin boosters such as 4-hydroxyisoleucine [Images] can be used at the end of a workout to phosphorylate tankyrase 1, making it active for this reaction. Furthermore, tankyrase 1 expression is known to be enhanced by niacinamide, so that in May for the first time I will be consciously opening telomere loops with tankyrase 1 before acting on them with telomerase expressed via the upregulation of hTERT mRNA levels using small molecule telomerase activators.

Boosting HGH levels for growth hormone telomerase activation
I also used whey protein, another HGH secretagogue added to arginine to kick HGH up as high as possible to get higher hTERT mRNA levels to support telomerase activity (7). However, unless I get behind heavier iron, nothing much will happen to my physical size from doing pushups, curls with light dumbells, and other light exercise, although my HGH levels are high. Creatine monohydrate is actually more anabolic than whey protein for supporting muscle growth and avoiding old age sarcopenia. I had to do 35 pull-ups with 100 lb chained on to get 19 inch arms back when I was really doing some bodybuilding. Creatine monohydrate could usually get me through sticking points, which is how I pressed 300 lb at 48. Creatine monohydrate also elevates IGF-1 levels and lead to 10% - 12% longer lifetime in experimental animals. Caloric restriction, however, ought to eventually produce a leaner, more photogenic look. I don't know why I find caloric restriction such a devilish challenge.

P16/Rb Inhibition
Carrots may taken daily to improve P16INK4A inhibition from retinol conversion to retinoic acid and to provide more retinoic acid obtained from beta carotene. Substantial daily bodybuilding exercise will further inhibit P16INK4A, promote exercise-induced telomerase activators for telomere maintenance, lower the LDL/HDL cholesterol ratio, improve lipid profiles, and elevate testosterone for reverse cholesterol transport to oppose the development of atherosclerotic plaque.

Costs seem to range between $110 to $200 dollars per month from discount drug stores and health food stores when astragalus root powder is the mainstay of the program. Ideally, a more expensive program reduces the amount of pill swallowing to be endured. Product B seems to cut the number of pills to for a 5 < |B| < 8 years/year rejuvenation program to 6 caps per day from 66 caps x 500 mg = 33 gr per day if NOW astragalus root is used. Using the readily available NOW astragalus root, a tolerable program uses 4 doses per day of 18 + 16 + 16 + 16 = 66 pills. However, things go better if GAIA astragalus root extract is also rubbed into the skin and scalp from green label GAIA Herbs astragalus root extract. Also, if you are free of atherosclerotic plaque with normal blood pressure and heartbeat and can work out with weights, things are more likely to work out, based on 14 telomerase activators from exercise therapy.

From test photos, I'd say I seem to be unsmiling because I have minor dental flaws I forget about that do require correction. I had to twist a front tooth to make it come in right as a child, and in some photos the shades make it seem to be a snaggle tooth after all. I had better see to carefully correcting everything in the dentition to emerge with a victorious grin. I unconsciously cover for it by not smiling much.
